Norvasc (amlodipine) is used with or without other medicines for high blood pressure (hypertension) treatment. Norvasc lowers high blood pressure and helps to prevent kidney problems, strokes and heart attacks. Norvasc active substance amlodipine is called a calcium channel blocker. Amlodipine works by relaxing blood vessels so blood can flow more easily. In several clinical trials, Norvasc has been safely administered with many medicines including: beta-blockers, warfarin, angiotensin-converting enzyme inhibitors, non-steroidal anti-inflammatory drugs, long-acting nitrates, sublingual nitroglycerin, antibiotics, oral hypoglycemic drugs, thiazide diuretics and digoxin. Norvasc and effect of other agents Sildenafil: A single 100 mg dose of sildenafil (Viagra, Kamagra or Silagra) in subjects with essential hypertension did not alter the pharmacokinetic parameters of Norvasc. When amlodipine and sildenafil were used together, each agent independently exerted its own lowering effect on blood pressure. Maalox (antacid): Combination of the antacid Maalox with a single dose of Norvasc had no significant impact on the pharmacokinetics of Norvasc. Cimetidine: Combination of Norvasc with cimetidine did not have effect to the pharmacokinetics of Norvasc. Grapefruit juice: Combination of 240 ml of grapefruit juice with a single oral dose of Norvasc 10 mg in 20 healthy volunteers had no important effect on the pharmacokinetics of amlodipine. Other agents and effect of Norvasc Ethanol (alcohol): Single and multiple 10 mg doses of Norvasc had no important effect on the pharmacokinetics of ethanol. Digoxin: Combination of Norvasc with digoxin taken by normal volunteers, did not change serum digoxin levels or digoxin renal clearance. Warfarin: Norvasc taken with warfarin did not change the warfarin prothrombin response time. Atorvastatin: Multiple 10 mg doses of amlodipine and 80 mg of atorvastatin taken together resulted in no significant change in the steady state pharmacokinetic parameters of atorvastatin. |
